Mechanism of action of Ethyl chloride:
Ethyl chloride is a condensed liquid under pressure. Because of its low boiling point (between 12° and 13° C) at atmospheric pressure, when it is sprayed onto the skin , it evaporates rapidly and thus cools the skin and produces freezing of superficial tissues , cold blocks nerve conduction resulting in insensitivity of peripheral nerve endings. This produces transient paralysis of cutaneous sensory nerve endings and local anesthesia.
Ethyl chloride creates an instantaneous cooling effect on the surface of the site by immediate evaporation of the product. The coldness created by the spray interrupts the body’s ability to experience pain.
This happens because the coldness decreases the nerve conduction velocity of the C fibers and A-delta fibers that make up the peripheral nervous system. Menthol :
It is a substance used as a topical application to induce a feeling of coolness as it has local anesthetic activity.
Mechanism of action of Menthol:
Menthol causes a tingling sensation and a feeling of coolness due to stimulation or activation ofTRPM8 (Transient receptor potential action channel subfamily M member 8) which is also known as the cold and menthol receptor1.
TRPM8 is an ion channel , upon activation it allows the entry of sodium ions (Na+) and calcium ions (Ca2+) to the cell that leads to depolarization and the generation of an action potential, leading to the sensation of cold.
